Phenotype
There is no obvious phenotype attached to the prt1 mutation. It can be assessed by PCR: primers CAG AGG AAG AGC AAG AAC GAG AAT and CCA CCT TCT GTT TAT CTA CAC give a band which can be digested with MnlI (recognition sequence CCTC) in the wild type, but not in the mutant (change to CCTT). Line 4H prt1 carries the prt1-1 mutation in the 4H background, i.e. with transgene. The transgene confers Hygromycin resistance, but in addition, as a consequence of the homozygous presence of the prt1 mutation, it confers resistance to methotrexate (0.1mg/l).
